Welcome to DMTDBThe classical view is that methylation of DNA at cytosines within cis-regulatory elements blocks recruitment of TFs. Recently many studies found that numerous TFs have methylation-dependent regulatory activity. However, a global landscape of DNA methylation mediated transcriptional dysregulations (DMTDs) across various cancer types hasn’t been portrayed. Here, we identified DMTDs systematically by performing an integrative analysis of trancriptome and methylome as well as regulatome and revealed that transcriptional dysregulations are prevalently affected by DNA methylation across 22 cancer types, involving 417 methylation sensitive TFs (MethTFs). Moreover, we classified 468 TFs in analysis into four categories according to the number of occurred cancer types: non-sensitive, cancer-specific, moderate and pan-cancer. DMTDB was built for users to search or download information of their interest in a flexible manner. For experimental researchers, we provided two modules to search: DMTD and MethTF. In the first module, users could query a factor associated DMTDs in a specific cancer type or in the global cancer background by submitting symbol or ID of interesting factor , and this platform will return a list of matched entries with detailed information about the identification and characterization. In the second module, users could get detailed information of a TF in a specific cancer type, including patterns and the target number of different patterns, by submitting symbol or ID of interesting TF. For computational researchers, all the datasets could be downloaded freely in the ‘Download’ page. Users could get instructions in the ‘Help’ page.
