To verify whether miR-34b and miR-34c directly target the 3-UTR of a-syn mRNA, we utilized a plasmid construct expressing full-length a-syn 3-UTR downstream of the firefly luciferase reporter gene. Co-transfection of miR-34b or miR-34c along with this reporter construct significantly decreased luciferase activity, but did not affect the control vector pGL3 devoid of thea-syn 3-UTR, indicating that miR-34b and miR-34c work through the 3-UTR of a-syn.
