Funtion Description For functional validation, genetic deletion of KDM4C by CRISPR-Cas9 was induced and compared to daunorubicin treatment as a positive control to induce cellular senescence. Here, we found induction of H3K36me, H3K9me3 and p21 (data not shown) along with significant increase in SA-betaGal staining in JAK2-mutated HEL cells in vitro.These findings indicate that inactivation of KDM4C reduces cell competition and proliferative capacity and induces cellular senescence in JAK2-mutated cells.
