Functional description
Eight miR-155 target sites with a bulge from nt 9 to 12 were cloned into the 3'UTR of the MS2 viral coat protein. We first investigated the functionality of the miR-155 sponge by the luciferase assay. In the presence of miR-155, luciferase levels of JARID2 3'UTR were reduced. With increasing amounts of miR-155 sponge, miR-155 function was inhibited and the repression of luciferase JARID2 3'UTR was abrogated
