Mechanism/Pathway Inflammation is considered as the first step in the pathophysiology of MS, although initiation mechanism is not well understood.There is increasing recognition that the disturbance of blood brain barrier (BBB) permeability and apoptosis pathway are associated with MS etiology.The Vaccinia Related Kinase 2 (VRK2) gene is located in 2p16.1 chromosomal region.VRK2 codes for a serine threonine kinase of the casein kinase 1 group.It has 9 transcripts due to alternative splicing and expressed in many tissues such as brain and blood.VRK2 participates in apoptosis regulating signaling pathways, immune responses, tumor cell growth, brain volume and structure.Due to the involvement of VRK2 gene in apoptotic pathway and apoptosis dysregulation in MS patients, it is most probable that it plays a regulating role in MS.
