| Result | Approximately 45% of the LEW. NR2 and LEWR1 displayed this relapsing–remitting pattern. Relapses were evident to a lower degree in LEW.NR3 and the parental LEW.N strain.Lymph node cells from rMOG(a.a.1-125)/IFA-immunized LEW.N, LEW.LR1, LEW.NR2 and LEW.NR3 rats responded to rMOG(a.a.1-125) stimulation in vitro with a relatively high number of IFN-g secreting cells. In contrast, there was no detectable T cell response in cultures derived from LEW and LEW. NR1 rats .In rats induced with the MBPGP63-88 peptide/CFA only LEW and LEW.NR1 rats, but not LEW.N, LEW.LR1, LEW. NR2 and LEW.NR3 rats, had detectable IFN-g responses upon restimulation with the immunizing peptide.We measured titers of antigen-specific antibodies in sera 12 days p.i.Susceptible strains raised significantly higher antibody titers than the resistant strains .Th1 (IgG2b) and Th2 (IgG1)-associated immunoglobulin isotypes were similarly affected in resistant and susceptible strains, demonstrating that there is no bias towards a Th2-like antibody response in the resistant strains.Immunization with recombinant myelin oligodendrocyte glycoprotein (a.a.1-125; MOG)/IFA induced EAE in strains expressing the MHC class II allele RT1.Bn , whereas strains expressing the RT1.Bl were resistant. In myelin basic protein peptide (MBPGP63-88)/CFA-induced EAE, RT1.Bl expressing strains were susceptible whereas strains expressing the RT1.Bn were resistant. High levels of antigen-specific IFN-g secreting lymphoid cells and antigen-specific serum IgG antibodies were only recorded in rats with an MHC class II allele that permitted MOG- or MBP-EAE, respectively. |
