Title EFFECTS OF VESNARINONE ON CYTOKINE PRODUCTION AND ACTIVATION OF MURINE MICROGLIA

Result In the CNS, TNFa is produced by astrocytes and microglia (4). Microglia may be the main producer of TNFIx, because they respond to produce TNFcr much faster than astrocytes and respond to much lower concentration of lipopolysacclharide (LPS) than do astrocytes (4). In this study, we examined the effects ofvesnarinone on TNFa production by microglia. The effects on the other functions of microglia such as proliferation, expression of major histocompatibility complex antigen, enzyme activity and superoxide anion formation were also examined.
Mechanism/pathway Tumor necrosis factor 01 (TNFcr) is considered to play a critical role in the development of various pathological processes in the central nervous system (CNS), such as neuronal degeneration, demyelination and gliosis. In order to search for agents which suppress TNFcx production in the CNS for future treatment of these pathological conditions, the effects of a synthetic oral inotropic agent, vesnarinone, on murine microglia were examined. Vesnarinone significantly suppressed TNFa production by microglia in a dose-dependent manner, without affecting their viability, enzyme activity or expression of the major histocompatibility complex.
