To analyse the expression of the miR-17-92 cluster, we performed real-time stem-loop RT-PCR of miRNAs in primary human astrocytic glioma tissue specimens and in non-neoplastic brain control tissues. Expression levels of miR-17-3p were significantly higher in primary Glioblastoma (GBIV) relative to non-neoplastic brain. Expression levels of miR-17-3p, miR-17-5p, miR-92a-1 and miR-106b were significantly higher in secondary GBIV relative to non-neoplastic brain. mir-19a and mir-19b were expressed at lower levels compared with the nonneoplastic brain. miR-17-5p, miR-19a and miR-106b showed a significantly higher expression in secondary GBIV specimens compared with astrocytoma grade II specimens.The CTGF gene was shown to be a target of miR-17-92 in glioblastoma spheroids by luciferase reporter assays. |
