the predominant consequence of p53 induction in this model system was widespread repression of miRNAs expression (17 p53-repressed miRNAs of 31 total p53 responsive miRNAs;P<0.05).stem loop quantitative RT-PCR (qRT-PCR) analysis in cells with activated p53; miR-25 and -32 were significantly down-regulated on treatment with Nutlin-3a in p53-activated U87 cells, whereas miR-34a, a direct target of p53, was up-regulated.miR-25 and -32 result in p53 accumulation by directly targeting Mdm2 and TSC1, which are negative regulators of p53 and the mTOR (mammalian target of rapamycin) pathway, respectively, leading to inhibition of cellular proliferation through cell cycle arrest. |
