miRNA profiles were generated for primary MB specimens, MB cell lines, CD133+NSCs and CD1332NPCs using qRT-PCR.clustering analysis based on the 50 most significantly differentially expressed miRNAs in both MB primary specimens and cell lines compared to CD133+NSCs was consistent with these findings.Significant upregulation of miR-935 was confirmed by qRT-PCR, with,11 000 fold up-regulation of miR-935 in pre-miR-935 transfected cells at 24 h compared to scrambled negative control cells.Significant down-regulation of three of the seven predicted miR-935 target genes was observed at the mRNA level Solute carrier family 5 member 3 (SLC5A3)(p = 0.0251), Hypothetical protein LOC9778 (KIAA0232)(p = 0.0004) and Zinc Finger AN1-type domain 6 (ZFAND6) (p = 0.0037). Furthermore, down-regulation of TBC domain family member 9 (TBC1D9) approached statistical significance (p = 0.064). |
